چکیده

Mastitis, a severe infectious disease among dairy cattle, is still a challenge for livestock treatment due to the increased infections by antibiotic resistant strains. Antimicrobial peptides could be the alternate for treating such drug resistant strains. In the present study, the whey proteins extracted from buffalo milk was subjected to enzymatic hydrolysis using trypsin, chymotrypsin, pepsin, papain and Proteinase K. Antibacterial assay carried out with these hydrolysates against Escherichia coli, Staphylococcus aureus, Streptococcus agalactiae and Streptococcus dysgalactiae isolated from mastitis milk showed that these hydrolysates have an inhibitory effect on their growth. Papain hydrolysates showed highest antibacterial activity against E.coli, Staphylococcus aureus, Streptococcus agalactiae and Streptococcus dysgalactiae. The fraction 4 (12 min fraction) of RP-HPLC purified papain hydrolysate showed highest antibacterial activity and that fraction has got Minimum inhibitory concentration of 100 μg/ml against E.coli and 150 μg/ml against Staphylococcus aureus, 100 μg/ml against Streptococcus agalactiae and 100 μg/ml against Streptococcus dysgalactiae.

Comparing the Activity and Thermal Inactivation Behavior of Lactoperoxidase in Iranian Cow and Buffalo Milk and Whey

Background: Lactoperoxidase (LPO) is one of the most heat-stable enzymes in milk and its inactivation has been proposed for monitoring thermal processes. The aim of this study was to provide information on activity and thermal inactivation behavior of LPO in Iranian cow and buffalo milk and whey. Methods: Sixty cow and buffalo milk samples were collected. The LPO activity was measured using ...

Antibacterial Effect of Bovine Lactoferrin Against Udder Pathogens

The antibacterial effect of lactoferrin (Lf) was tested on isolates of Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), and coagulase-negative staphylococci (CNS) as well as on Pseudomonas aeruginosa (P. aeruginosa) and Klebsiella pneumoniae (K. pneumoniae), originally isolated from bovine mastitis. Concentrations of Lf used were 0.67 mg/ml, 1.67 mg/ml, and 2.67 mg/ml. Growth of u...

Actinopyga lecanora Hydrolysates as Natural Antibacterial Agents

Actinopyga lecanora, a type of sea cucumber commonly known as stone fish with relatively high protein content, was explored as raw material for bioactive peptides production. Six proteolytic enzymes, namely alcalase, papain, pepsin, trypsin, bromelain and flavourzyme were used to hydrolyze A. lecanora at different times and their respective degrees of hydrolysis (DH) were calculated. Subsequent...

Antimicrobial Peptides Derived from Goat’s Milk Whey Proteins Obtained by Enzymatic Hydrolysis

In this study the bacterial growth inhibitory activity of peptide fragments produced from goat’s milk whey proteins by enzymatic hydrolysis using trypsin, ficin and a combination of both was investigated. Goat’s milk whey proteins were isolated and subjected to enzymatic hydrolysis and peptides were purified by ultrafiltration followed by reverse-phase high-performance liquid chromatography (RP...

Antibacterial peptides derived from caprine whey proteins, by digestion with human gastrointestinal juice.

Peptides in caprine whey were identified after in vitro digestion with human gastrointestinal enzymes in order to determine their antibacterial effect. The digestion was performed in two continuing steps using human gastric juice (pH 2·5) and human duodenal juice (pH 8) at 37°C. After digestion the hydrolysate was fractionated and 106 peptides were identified. From these results, twenty-two pep...
